BVRA (biliverdin reductase A) was knocked out of the mouse proximal tubule cells (MCT) using sgRNAs inserted into the GuideIt-zsGreen plasmid. The plasmid was transfected and clones were selected via FACS sorting for fluorescence. Knockout was determined by qPCR and Western blot analysis. Studies have shown the loss of BVRA is associated with fat accumulation in livers of mice models. Therefore, BVRA Stable Knockout MCT Cell Line is useful to study the effects of BVRA and its association with obesity, increased lipid availability, and lipotoxicity.
