The E. coli CRISPR Knockout Teaching Kit allows students to gain hands-on laboratory experience utilizing CRISPR - a revolutionary gene-editing technique. In this robust experiment, the student performs a gene knockout of a genomically-encoded chloramphenicol-resistance cassette employing CRISPR and lambda red recombineering. This results in E. coli that is now sensitive to the antibiotic chloramphenicol – a phenotype that can be easily screened for. The experiment is carried out in a safe laboratory strain of E. coli (BSL-1) and uses standard microbiological/molecular biology reagents and equipment allowing for ease of implementation in most teaching labs.

One unit is defined as the amount of E. coli DNA Polymerase I that catalyzes the incorporation of 10 nmol of dNTP into acid insoluble material in 30 minutes at 37°C using poly(dA-dT):poly(dA-dT) as a template:primer.
