The GALE gene encodes UDP-galactose-4 epimerase which operates in the Leloir pathway, the primary pathway for galactose metabolism. This pathway is critical for the generation of UDP-sugars and catalyzes UDP-glucose to UDP-galactose and UDP-N acetylgalactosamine to UDP N-acetylgalactosamine which serve as precursors for O-linked glycosylation and as the major metabolism pathway for galactose. As this cell line is derived from a human source, it may be useful in the study of glycosylation and how it is affected during instances of disease.

1) Confirmed CRISPR-Cas9 presence by GFP examination (Figure 1). 2) Confirmed GALE knockout through western blot analysis and sequencing (Figure S1). Tested O-glycosylation levels through enzymatic activity on the substrate SIVmac239 gp120 via Western Blot and SDS gel analysis (Figure 3)
