Propagation:

The base medium for this cell line is Prigrow II medium available at abm . To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10% and Penicillin/streptomycin to a final concentration of 1%.
Carbon dioxide (CO₂): 5%, Temperature: 33.0°C. The functional expression of the SV40 antigen is induced by culturing the cells in vitro at a temperature permissive (33°C). At a non-permissive temperature (37°C-39°C), the cells cease to proliferate and revert to differentiated phenotype.
abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.

Quality Control:

1) Northern blot performed to analyze presence of activin βA , follistatin and PLP-B mRNA; 2) RT-PCR analysis of progesterone receptor, activin βA, activin βB, and activin receptor II mRNA was completed with L19 ribosomal protein mRNA as control.
