Deoxyribonuclease I (DNase I; EC 184.108.40.206) is a non-specific endonuclease that catalyzes the cleavage of phosphodiester bonds in single/double-stranded DNA, chromatin, and RNA-DNA hybrids. DNase I cleaves DNA to release di-and/or oligonucleotide (5’-phosphorylated and 3’-hydroxylated) end-products.
One unit is defined as the amount of Dpn1 required to digest 1 µg of dam methylated pBR322 DNA in 1 hour at 37°C in a total reaction volume of 50 µl.
