When cells undergo death triggered by chemotherapeutic agents, the BCL-2-family-dependent mitochondrial apoptotic pathway is activated. By activating BAX and/or BAK, one of the BCL-2 subgroups, BH3-only proteins cause cytochrome to release from the mitochondria. shRNAs were introduced specific for BAK and BAX in HN12 cells so their roles can be studied. The results suggest that both BAX and BAK are required for apoptosis induced by simultaneous inhibition of BCL-2.
